Use with caution and consider dose adjustment in animals with renal impairment. Not for use in rabbits, guinea pigs, hamsters, or other small herbivores, in which penicillins cause fatal disruption of the gut flora. Drools CaninGuard is a prescription injectable antibiotic for dogs and cats combining Amoxicillin, a broad-spectrum aminopenicillin, with Sulbactam, a beta-lactamase inhibitor. Many of the bacteria that cause wound, urinary, and respiratory infections defend themselves by producing beta-lactamase enzymes that destroy penicillins before they can work. Sulbactam blocks those enzymes, restoring Amoxicillin's activity against organisms that would otherwise be resistant to it. The injectable route delivers therapeutic blood concentrations quickly, making it well suited to acute infections, post-surgical cover, and patients that cannot reliably take oral medication. Available in 300mg and 600mg strengths for weight-appropriate dosing.

Composition & Mechanism of Action

• Amoxicillin — aminopenicillin, beta-lactam class, bactericidal. Amoxicillin binds to penicillin-binding proteins in the bacterial cytoplasmic membrane and blocks the transpeptidation reaction that cross-links peptidoglycan strands in the cell wall. Without those cross-links the wall cannot withstand internal osmotic pressure, and the growing bacterium lyses. Because peptidoglycan walls exist only in bacteria, the margin of safety in the treated animal is wide. The aminopenicillin structure gives Amoxicillin broader Gram-negative reach than the natural penicillins.
• Sulbactam — irreversible beta-lactamase inhibitor. Sulbactam has little antibacterial activity of its own; instead it acts as a suicide substrate, binding covalently and irreversibly to bacterial beta-lactamase enzymes and permanently inactivating them. Those enzymes would otherwise hydrolyse the beta-lactam ring of Amoxicillin and render it useless. By sacrificing itself, Sulbactam protects Amoxicillin and restores its activity against beta-lactamase-producing strains of Staphylococcus, Escherichia coli, Klebsiella, and Proteus.
• Synergistic effect — the pairing gives faster and irreversible beta-lactamase inhibition and a broader effective spectrum than Amoxicillin alone, covering Gram-positive and Gram-negative aerobes as well as anaerobes commonly involved in bite wounds and abscesses.
